Birefringence polarized light microscopy
WebSep 1, 2024 · Maize starch granules are polygonal with a size of 11.7 ± 2.1 μm. Sweet potato starch granules with an oval-shape have an average size of 15.2 ± 3.8 μm, larger than both rice and maize starches. Because the resolution for an optical microscopy is usually around 200 nm, the obtained size by DFM is accurate. WebThis condition is known as birefringence. Polarized light microscopy produces images by utilizing the interference of the split light rays as they re-assemble along the same optical path. The split rays travel at different speeds through the specimen and one ray is retarded with respect to the other. This optical condition is known as retardation.
